When setting a new hotkey on something, after pressing the "Ok" button on the "Cheat Engine settings" window, it will then do the action that was assigned to that key.

For example if you were to set a hotkey for 'Next Scan-Changed Value' to the "End" key, after pressing "Ok" it will rescan for changed values.
