With a program, I'm sure. I don't think you can do it physically though. Correct me if I'm wrong. It would take a special hard drive with the feature.

Tried bitlocker on my Vista Ultimate and this is what I've found out.
- You will need a TPM module for it to work, but you can change bitlocker to use a USB flash drive.
- It takes a long time to get the drive encrypted
- When the encryption is done, it will give you a random key, this is used as a backup in case you lost or broken your USB flash drive.
- If it's setup correctly, it will not boot without the USB key
- If USB key is lost or broken, it will ask for than random generated code.
There are hardware based encryption drives, Seagate make them.
You can also use 3rd party software like TrueCrypt
