First off make sure your resolution is low. (800 by 600)
graphics option are low
textures on lower quality
some games (minecraft) render distance on a medium-low setting
Scan with AVG PC tuneup, this tool is NOT an antivirus.

Other Hints:
1. Keep large files off your desktop! it slows it down
2. Clear the recycling bin (dam windows)
3. Detfragment your drive regularly
4. scan with an antivirus (i suggest avg's antivirus)

Windows does load the desktop another way that it uses just a "regular" folder. Because it is visually loaded for showing you nice thumbnails and such, it can take a long time indexing everything on there. Point is that that takes time when starting windows, thus slowing it down. Also, when aero is enabled, it is stored in the graphical memory, even when in full-screen mode. You can prevent this from happening by checking the box under the second tab (in the properties screen) of your game, named "Disable desktop composition". This will temporarily shut down aero, for as long as that program runs.
Also note: youtube video decoding acceleration (which is auto-enabled if your drivers support it) takes quite a lot of memory of your graphics card. I advise against any browser screens while playing games, even with better cards.
Example of the latter:
I have a GTX 260 with 896MB memory. I have 4 youtube videos open, and running dual screen windows 7 with aero. I currently use up 300MB's of that memory. When I close the browser, it goes down to 50MB in use.
