These are my specs:
E8400
4GB Ram
Radeon HD4850
1TB HD
EP45-UD3P motherboard
MODULAR RX-630A 630W power supply
I am using a Razer Piranha headset, and my computer does not recognize that I have a microphone. I plugged the pink jack in the pink hole.
I have three different types of mixer devices:
Realtek HD Audio output
Realtek HD Audio input
Realtek HD Digital input
I tried checking all the boxes in all 3 settings.
I am also unable to set my Sound device to "headset" like on my old computer.

And what driver do you have for the Realtek audio? The full program has the interface thing where you can choose what everything plugged in goes to.

It would probably have to do with "Audio Input" since you plugged it in the pink jack. If the Realtek program doesn't help, I don't know where to go from there. Just got here from flying.

Mic-In is correct, but make sure your mic is not muted in the volume control panel.
Try changing your audio standard to AC-97 in the bios.

Look at your motherboard's manual on what key to press for accessing the bios and look somewhere for audio device.
Default should be HD Audio, change that to AC-97, save and exit.

Download the latest driver for your Realtek HD sound card from Realtek's website.
The HD sound driver is universal so no need to worry about what motherboard you have.
Before doing that, make sure your
1. Mic is not muted!
2. It's actually turned on
3. The volume is turned up.
4. Open sound recorder and record something to see if mic works.
